MudPIT (Multidimensional Protein Identification Technology) or multi-dimensional
LC/MS/MS
MudPIT or multi-dimensional LC/MS/MS analysis is a powerful tool for protein
identification in a complex mixture (such as a cell extract, isolated organelle
fraction, or a mix of 10 or more proteins) that would overwhelm the normal
reverse phase LC/MS/MS analysis. Usually, other technologies, such as 2D gel
electrophoresis, are used for separating complex mixtures of proteins prior to LC/MS/MS
assay. We are using custom-packed LC column with
strong cation exchange (SCX) material packed back-to-back with the standard
reversed phase (RP) material and LC-MS/MS technology to carry our
Multidimensional Protein Identification, which has been successfully used for
Yeast proteome.
